MOTS-c (Mitochondrial ORF of the 12S rRNA Type-C) is a 16-amino acid mitochondria-derived peptide encoded by mitochondrial DNA, representing a novel class of intracellular signaling molecules. It activates the AMPK pathway and regulates the folate cycle and de novo purine synthesis under metabolic stress. During cellular stress, MOTS-c translocates to the nucleus and directs nuclear gene expression to restore metabolic homeostasis. Research has demonstrated its roles in improving insulin sensitivity, enhancing glucose uptake, reducing oxidative stress, improving mitochondrial bioenergetics, and modulating inflammation in aging, cardiovascular, metabolic, and orthopedic disease models.
